# Engineered Gut Bacteria Turned Into Drug Factories Against Pancreatic Cancer

> University of Chicago scientists engineered a gut probiotic to deliver immune-stimulating therapy straight into pancreatic tumors, slowing their growth in animal studies.

University of Chicago researchers engineered a common probiotic, Bifidobacterium longum, to act as a microscopic drug factory inside pancreatic tumors. The modified strain, called BifidoSumIL-2, produces a tumor-targeted form of interleukin-2 that activates cancer-fighting CD8+ T cells directly at the tumor site.

The bacterium naturally thrives in the low-oxygen environment inside tumors while being cleared from healthy, oxygen-rich tissue, letting the therapy concentrate where it's needed and avoid flooding the whole body with immune stimulants. In mouse models, the treatment slowed pancreatic tumor growth on its own and worked even better when paired with chemotherapy, radiation, or anti-PD-L1 immunotherapy.

Pancreatic cancer has resisted most immunotherapies because its tumors create a "cold" microenvironment that blocks immune cells from mounting an attack. The study, led by Ralph Weichselbaum and Mark Mimee and published in Science Advances, has not yet been tested in humans; next steps include evaluating long-term safety and whether the bacteria could one day be given orally.
